There are two competing approaches to the PoW puzzle that were decided upon in advance. The first is that chains should be long. This is because miners have to search for those long chains and can’t make a profit by mining shorter chains.
The second is that it should be difficult to reverse engineer the block chain and come up with the hash that creates it. This prevents 51% attacks.
The spacing is a matter of engineering compromise between those two ideas. With 1 block per minute, 5 minutes is 2,000 blocks. With a difficulty increase every 10 minutes, it takes 1,000 blocks to clear the difficulty bar.
