The interaction between dietary protein and starch concentrations has not been evaluated in regard to regulation of liver gene expression in broiler chickens. Therefore, genes involved in pathways of energy metabolism and intermediary metabolism as well as growth and regulation of lipid metabolism were studied in broiler chickens fed 1 of 5 protein/starch ratios at 21 to 49 days of age. Dietary protein content was manipulated by using a corn-wheat pellet diet formulated to contain 14%, 17%, 19%, 20%, and 22% crude protein. Starch content was manipulated by using diets containing 45% corn, 35% corn, and 50% corn by replacing starch with dextrose. The diets were fed either ad libitum or at a restricted level of 8% of dietary intake. Liver microsomal cytochrome P-450 content and serum insulin-like growth factor-I (IGF-I) were used as markers of protein status and size of the liver. The relative liver content of cytochrome P-450 and the concentration of hepatic IGF-I were significantly reduced in birds fed the 17% protein diet, with no difference in the content of these proteins in the livers of birds fed the 21% protein diet. Therefore, dietary restriction of dietary protein did not result in a significant change in the activity of cytochrome P-450, and hepatic cytochrome P-450 protein abundance was reduced in birds on the 17% protein diet. The concentration of hepatic IGF-I was significantly greater in birds fed the 8% protein diet than in birds on the 21% protein diet. Liver cytochrome P-450 content and serum IGF-I concentration were not significantly different among diets containing 14%, 17%, 19%, 20%, and 22% dietary protein. Liver weight was significantly greater in birds fed the 8% protein diet than in birds on the 21% protein diet. Liver weight was also significantly greater in birds fed the diet containing 45% corn, 35% corn, and 50% corn. Microsomal cytochrome P-450 and the liver:body weight ratio were linearly related to liver weight, and genes in these pathways were differentially regulated by protein/starch ratio.
